The course will focus on the importance and role of RNA processing alteradons in human disease, from simple monogenic diseases to complex pathologies such as cancer and neurodegeneration. The course will be divided in two parts: the first part ( 20 hours) will be theoretical and will describe the role played by RNA alterations in human disease. The second part (8 hours) will take advantage of the Teaching Lab in ICGEB to introduce students at basic laboratory RNA manipulation techniques such as extraction, quality control, detection, and amplification..In particular, the first part will provide an Introduction to RNA and its application to Molecular Medicine. This will be followed by an explanation of the role played by RNAs in our metabolism (mRNAs, siRNAs, IncRNAs, etc.) and in normal development/metabolism (its origin, regulation, and consequences). Then, the students will receive a brief introduction with regards to the role played by RNA in disease (impact on human health) and in particular on the recent development of RNA-based therapeutic strategies to rescue aberrant splicing events (antisense oligonucleotides, small molecules, screening procedures, etc.). The e, on the other hand, will consist of a few practical lessons that will take place in the teaching lab at the university of Trieste. These lessons will consist making the students isolate total RNA from culture cells, perform CDNA synthesis for specific factors, and use RT-PCR and RT-qPCR approaches to detect the effect of nucleotide substitutions that can affect RNA processing evenD such as pre-mRNA splicing.
